How it works
Agomelatine belongs to a class of drugs called melatonergic antidepressants. It binds to the melatonin receptors (MT1 and MT2) to promote their activity and serotonin receptors (5HT2C) to block their activity in brain, thereby improving sleep efficiency, normalizing day-to-day rhythms involved in normal functioning of brain, all of which causes improvement in symptoms of depression.
Dosage
Always follow the instructions of doctor regarding the dosage and duration.
Adults (>18 years): 25 mg at bedtime, increased if necessary after 2 weeks to 50 mg at bedtime.
Common side effects
Anxiety, headache, sleepiness (somnolence), pulsating headache often on one side of the brain (migraine), lack of sleep (insomnia), dizziness, nausea, diarrhea, constipation, stomach (abdominal) pain, vomiting, excessive sweating (hyperhidrosis), back pain, unusual tiredness (fatigue), increased liver enzymes in blood.
Who should not take
Patients who are allergic to agomelatine or to any of its ingredients.
Patients with hepatic impairment.
Patients taking other antidepressants (?uvoxamine) or an antibiotic (ciprofloxacin).
Patients with sugar intolerance.
Elderly patients with major depressive episodes with dementia.
Children under 18 years of age.
